 Obesity is the major risk factor for cardiovascular disease, but its mechanism is still not clear. Researchers have proposed two possible hypotheses on obesity-induced diseases: inflammation and increase of oxidative stress. The study of obesity mechanism depends on the study of obesity related bio-markers, and it is very important to find the optimal bio-markers to improve the accuracy of cardiovascular diseases diagnosis.

The common used bio-markers are inflammation related bio-markers, for instance, IL6, Adiponectin, Leptin, etc, and oxidative stress related bio-markers, for instance, oxLDL, PPARg, MCP1, etc. Cloud-Clone Corp. has developed a series of quantitative ELISA kits for the detection of obesity related bio-markers, these products include kits for detection of 30 kinds of different bio-markers in early obesity (Fig. 1), and a series of recombinant proteins and antibodies corresponding to these bio-markers. Interleukin 6 (IL6) is a kind of inflammatory marker, whose main function lies in adjusting inflammatory signals and balancing of lipid by regulating the metabolism of lipid and expression of C reactive protein (CRP). As the expression of IL6 is very low in normal human tissue, Cloud-Clone Corp. had developed high sensitive ELISA kit for IL6 detection (HEA079Hu), the detection sensitivity of this kit had been improved greatly compared to the regular kit.

Adiponectin (ADP) is exclusively expressed in adipose tissue, it is a protein similar to complement factor C1q and some collagen molecules. Cloud-Clone Corp. have developed specific ELISA Kit (SEA605Hu) and CLIA kit (SCA605Hu) for ADP detection. Biotin and FITC-labeled antibodies with high quality are also provided (PAA605Hu71 / PAA605Hu81).

Leptin is produced by adipose tissue and heart. It can regulate the production of cytokines such as IL-6 and TNF-a, and it could also cause hypertrophy of smooth muscle cells. Increasing evidences support that leptin is an optical bio-marker to predict the risk of cardiovascular disease. So, Cloud-Clone Corp. had developed specific kit for in vitro quantitative measurement of leptin in various of human samples (SEA084Hu).

OxLDL is mainly produced by oxidization of low density lipoproteins in endothelial cells and macrophages, it could be accumulated on arterial walls by macrophages, and this process strongly contributes to the development of atherosclerotic lesions. Based on the native oxLDL purified from the tissues of human, mouse, rat, rabbit, etc, Cloud-Clone Corp. had developed a series of quality guaranteed ELISA kits for oxLDL detection (SEA527Hu /SEA527Mu / SEA527Ra / SEA527Rb).

The function of Peroxisome Proliferator Activated Receptor Gamma (PPARγ) mainly exists in adipogenesis and sensibilization of insulin. We had expressed two different recombinant protein of PPARγ (RPA886Hu01 / RPA886Hu02) , which contain different fragment with aboudant epitopes, through epitope prediction and analysis. RPA886Hu01 is the C terminal fragment of the human PPARγ, and RPA886Hu02 is the N terminal fragment of the human PPARγ.

Monocyte chemoattractant protein 1 (MCP-1) plays an important role in the recruitment of monocytes to positions of injury and infection. It is reported that TNF-a can induce MCP-1 expression in murine adipocytes, the kit of SEA087Mu could be used in this kind of researches. And researchers had found that MCP-1 is highly expressed in macrophages and endothelial cells of atherosclerotic lesions, and this phenomenon is positively associated with obesity and coronary artery disease in human. Biotin-Linked Antibody to MCP1 (PAA087Hu71)can be used for experiment of western blot and IHC.

In addition to these six major indicators, the expression levels of angiotensin converting enzyme (ACE), adipose differentiation-related protein (ADRP), IL10, IL1Ra, IL18, TNFa, and plasminogen activator inhibitor 1 (PAI1) in the entire family of obesity susceptible people are different from the normal ones.

The accumulation of fat will lead to the change of expression of inflammation and oxidative stress related biomarkers. The expression changes of these proteins will result in the expression changes of obesity related bio-markers, in return, and these interaction will finally lead to cardiovascular disease.
