Secreted frizzled-related protein 5 (SFRP5)-the target of Obesity Therapy

In recent years, obesity has increased dramatically. The metabolic diseases caused by obesity, including hypertension, coronary heart disease, high cholesterol, diabetes, fatty liver and gallbladder disease, cholelithiasis, etc. show a significant increase, obesity has gradually become one of the major health problems world widely.

It is reported that the adipose tissue, as a secretary organ, can secrete proinflammatory and anti-inflammatory adipocytokines. These factors are beneficial to health. And the disorder of adipocytokine secretion may be the main reason for human obesity.

Secreted frizzled-related protein 5 (SFRP5), belonging to secreted frizzled related protein (SFRP) family members, is a novel anti-inflammatory adipocytokines. SFRP5 can competitively bind to Wnt5a protein to inhibit Wnt signaling pathway and the activation of some critical inflammatory cells including macrophages in adipose tissue. Based on this, the secretion of inflammatory cytokines will be reduced. Since SFRP5 is regarded as a linkage of obesity and other metabolic diseases, further research of the interaction between SFRP5 and signaling molecules in wnt signaling pathway may lead to new ideas for the treatments of obesity and obesity-related metabolic diseases.

In view of this hotspot, Cloud-Clone Corp. has developed a series of SFRP5 recombinant proteins, antibodies and quantitative detection kits and other related products. The process of specific products is introduced as follows.

At first, Cloud-Clone Corp. uses a high-flux expression system to express recombinant protein of SFRP5. The cDNA fragment was linked with the self-established E.coli expression vector pUSCNK-1 by traditional double enzymes restriction and ligation after verified by DNA sequencing. Then the recombinant vector was used for protein expression. Subsequently, we got a recombinant protein of SFRP5 (with a his-tag) by IPTG induced expression. After purified by Nickel column, recombinant proteins with high purity (RPC842Hu01, RPC842Hu02, RPC842Mu01, RPC842Mu02, RPC842Ra01) were obtained. Take the recombinant human SFRP5 (RPC842Hu01) for example, SDS-PAGE test is shown in Figure 1.

Then we immunized mice by purified SFRP5 (purity>95%) and obtain large quantities of monoclonal antibodies by hybridoma technique, mouse anti-human SFRP5 (MAC842Hu22) and mouse anti-rat SFRP5 (MAC842Ra22). In addition, we also immunized rabbits to prepare polyclonal antibodies, such as polyclonal antibody to human SFRP5 (PAC842Hu01 & PAC842Hu02), polyclonal antibody to mouse SFRP5 (PAC842Mu01 & PAC842Mu02) and polyclonal antibody to rat SFRP5 (PAC842Ra01). All these antibodies were affinity purified, the purity and specificity of antibodies were verified by Western Blot. Take the mouse anti-human SFRP5 (MAC842Hu22) as an example, the results is shown in Figure 2.
