One Hot Spot in Cardiovascular Diseases-ANGs

It is reported by World Health Organization (WHO) that cardiovascular disease has become the leading cause of death. From the statistical data of 2008, death caused by cardiovascular disease is 30% of global death toll, and it is predicted that the ratio will increased by 18% in 2030. Due to the significant impact of this diseases on human being, researchers all over the world had pay persistent attention on it. Recently, lots of reports, which are about a group of peptides, angiotesin (ANG) , are published. Regarding to these peptides, our company had developed related reagents, so, they could be used by researchers to facilitate their research.

ANGs is a group of hormone consist of oligopeptides, they are derived from the precusor, angiotensinogen (AGT), which synthesized by liver, and degraded through renin and lots of enzymes. ANGs play an important role in vasoconstriction, blood pressure elevation and adrenal hormone releasing. The process of ANGs’ formation is shown in Figure 1, all the steps are completed in renin-angiotensin system (RAS), and the final products include peptides with five to ten amino acids.

From figure 1, it is found that there are only minor difference, eg: 1-2 amino acids, between each ANG peptide. The sequence of these peptides are shown in Figure 2. So, the most important point for ANGs detection kits preparation is the specificity. In R&D processes of the detection kits, we had prepared plenty of monoclonal and polyclonal antibodies, and focused on the specificity of them to exclude cross-reactivity. Moreover, as ANGs is a group of peptides with low molecular weight, which means each of the peptide does not possess several epitope, competitive inhibition detection method is applied in the detection kit.

Currently, several detection kits are pushed into the market, including Angiotensin I (AngI) (CEA811Hu), Angiotensin 1-9 (Ang1-9) (CET844Hu), Angiotensin 1-7 (Ang1-7) (CES085Hu), Angiotensin II (AngII) (CEA005Hu), Angiotensin III (AngIII) (CEC312Hu) and so on. We can see the specificity of the kits from one example, ELISA kit for Ang1-7 (CES085Hu), the data of cross-reactivity between this kit and AngI/Ang II is shown in Figure 3. From the data, we can find that, there is no significant cross-reactivity between CES085Hu and AngI/AngII.
