Receptor Interacting Serine Threonine Kinase 1 (RIPK1)
Two cell surface cytokine receptors, FAS and tumor necrosis factor (TNF) receptor, trigger apoptosis by natural ligands or specific agonist antibodies. Both receptors contain a conserved intracellular death domain. Using a yeast 2-hybrid screen with the cytoplasmic domain of FAS as bait, Stanger et al. (1995) isolated partial cDNAs encoding a receptor-interacting protein which they designated RIP. They used the partial cDNAs to isolate mouse cDNAs corresponding to the entire Rip coding region. Hsu et al. (1996) isolated a full-length human RIP cDNA. They reported that the predicted 671-amino acid RIP protein contains an N-terminal protein kinase domain, a C-terminal death domain, and a unique internal region that they called the intermediate domain.
