Advantages of Competitive Inhibition ELISA Comparing to HPLC--for Quantitative Detection of Small Molecules

Cloud-Clone Corp.

Small Molecules: Generally, small molecule refers to chemical compounds with  low molecular weight (MW), lower than 1000 Dalton (Da). Different small molecules play distince roles in the pathology of clinical and scientific field, for instance the level of some hormones such as estron can be used as a biochemical indicator in a wide variety of infectious and noninfectious diseases.

As a kind of typical hapten, small molecules lack immunogenicity which can trigger immunized animal to produce specific antibody. Thus, it is difficult for us to detect them quantified by conventional double-antibody ELISA method. However, as the development of molecule-coupling technology, plus our own modified structural modifying and coupling processes, our small molecule-specific ELISA kits are increasing being used in the scientific fields.

Competitive Inhibition Method: Due to unique structural characteristics of small molecules, it generally has no more than one epitope, thereby double-antibody sandwich method is not suitable for small molecule detection. So, we used the competitive inhibition method to establish small molecule-specific ELISA kits.

Comparing to conventional small molecule specific-detection method——high performance liquid chromatography (HPLC), the advantages of ELISA for quantitative detection of small molecules are shown below:

Table 1. Comparison of ELISA and HPLC in small molecules detection

Items HPLC ELISA
Sample Volume 10-100μL 50μL
Sample Characteristic High Moderate
Experimental Time/Sample90 samples/plate 20-45 min < 2 min
Sample Quantity 1 sample/tube Moderate
Equipment Price High Moderate
Detection Price/Sample High  Moderate

General Kit: as small molecules show the same structure in all species, i.e., one kind of small molecule specific kit can recognize the molecules derived from different species. This kind of kits is also been called general kits.

Currently, small molecule detection kits we developed has captured the attention of the public and the scientific community alike, including hormone such as estron (CEB003Ge), testosterone (CEA458Ge); amino acids and vitamins, e.g, Arginine (CEB938Ge), vitamine B1 (CED053Ge), etc.; lipopolysaccharide (CEB526Ge), abscisic acid (CEB218Ge) and so on.
